- Levy County was established on March 10, 1845, from a segment of Alachua County.

- Sumter County was established on January 8, 1853, from a segment of Orange County.

- Liberty County was established on December 15, 1855, from a segment of Gadsden County.
- Lafayette County was established on December 23, 1856, from a segment of Madison County.
- Taylor County was established on December 23, 1856, from a segment of Madison County.
- Bradford County was established on December 21, 1858, from segments of Columbia County and Alachua County.
- Suwannee County was established on December 21, 1858, from a segment of Columbia County.
- Clay County was established on December 31, 1858, from a segment of Duval County.
- Baker County was established on February 8, 1861, from a segment of New River County (Bradford County).
- Polk County was established on February 8, 1861, from segments of Brevard County and Hillsborough County.
- Osceola County was established on May 12, 1887, from segments of Orange County and Brevard County.
- Lee County was established on May 13, 1887, from a segment of Monroe County.
